Tris base is widely used in acute metabolic and respiratory acidemia. It is an alkaline buffer that has a good buffering effect on metabolic acidosis and enzyme activity reactions. Tris is commonly used as a biological buffer, often formulated with pH values of 6.8, 7.4, 8.0, 8.8. Its structural formula and pH value vary greatly with temperature. Generally speaking, for every one degree increase in temperature, the pH value decreases by 0.03. Tris is widely used in the preparation of buffer solutions in biochemical and molecular biology experiments. For example, the commonly used TAE and TBE buffer solutions (used for nucleic acid dissolution) in biochemical experiments require the use of Tris; Due to its amino group content, it can undergo condensation reaction with aldehydes. The pH of Tris alkali aqueous solution is around 10.5. Generally, hydrochloric acid is added to adjust the pH value to the desired value to obtain a buffer solution with that pH value. However, attention should also be paid to the effect of temperature on the pKa of Tris. Due to Tris buffer being a weakly alkaline solution, DNA is deprotonated in such a solution, thereby improving its solubility. People often add EDTA to Tris hydrochloric acid buffer to make "TE buffer", which is used for DNA stabilization and storage. If the acid solution used to adjust the pH value is replaced with acetic acid, a "TAE buffer" (Tris/Acetate/EDTA) is obtained, while replacing it with boric acid results in a "TBE buffer" (Tris/Borate/EDTA). These two types of buffers are commonly used in nucleic acid electrophoresis experiments. TAE and TBE, which are composed of Tris, are the most commonly used reagents for DNA electrophoresis. TE (pH 8.0) is mainly used to dissolve DNA (TE is a combination of Tris and EDTA). Tris buffer is not only widely used as a solvent for nucleic acids and proteins, but also has many important applications; It is used for protein crystal growth under different pH conditions. The low ion strength of Tris buffer can be used for the formation of intermediate fibers in the lamin of C. elegans, and Tris is also one of the main components of protein electrophoresis buffer. In addition, Tris is also an intermediate in the preparation of surfactants, vulcanization accelerators, and some drugs, and it is often used as a titration standard.
